角毛壳菌CH-1产生的抗真菌活性化合物的纯化和鉴定

Purification and Structure Elucidation of Antifungal Metabolite from Chaetomium cupreum CH-1

摘要: 对角毛壳菌CH-1进行液体发酵,以辣椒炭疽病菌为指示菌,研究发酵液的理化性质,以减少提取过程中活性化合物的失活,结果表明发酵液在60 ℃以内,pH 3~5之间活性稳定,乙酸乙酯对活性化合物萃取效果最好。采用活性跟踪分离的方法,运用萃取、正相硅胶层析、结晶、重结晶等技术从发酵液中分离纯化得到纯品。通过紫外光谱、红外光谱、质谱、核磁共振谱等波谱学手段鉴定其结构,确定化合物为卵孢菌素,其在防治植物病原真菌方面具有广泛的应用前景。

Abstract: Chaetomium cupreum CH-1 was fermented in liquid culture. To keep the activity of the antifungal compounds during the processing, we studied the physical and chemical properties of the broth using Colletotrichum capsici as bioassay indicator. It indicated that the broth would keep activities while the temperature within 60 ℃ and the pH values from 3 to 5, and the active compounds would be extracted effectively with the ethyl acetate. Purification of the active compound in the broth of the strain was performed using a combination of extraction, flash column chromatography, crystallization and re-crystallization. The active compound named oosporein was elucidated by UV-Vis, IR, HR-ESI-MS and NMR spectra and had a broad application prospect in prevention and control of plant pathogenic fungi.
